Juglans regia L. fruit pellicle extract-based bioreduction of silver nanoparticles: Structural features and in vivo therapeutic effects against ethanol-induced peptic ulcers

Abstract

Context: Peptic ulcers are a challenging problem. Silver nanoparticles (AgNPs) play a substantial role in bioactivity and have distinct characteristics. There are several biomedical uses for bioreduction nanotechnology. Historically, traditional healthcare has employed Juglans regia L.

Aims: To evaluate how well fruit pellicle extract-derived bio-reduced silver nanoparticles (AgNP/FP) can treat peptic ulcers.

Methods: Ultraviolet spectra, dynamic light scattering, transmittance electron microscopy, and Fourier transform infrared were used to characterize AgNPs and AgNP/FP. AgNPs and extracts were compared to omeprazole in ethanol-induced stomach ulcers in mice. Six groups of 36 mice were created at random. AgNPs were given orally to the experimental groups, using 300 mg/kg as a dose. The reference proton pump inhibitor, omeprazole, was employed as a control.

Results: As evidenced by a falling ulcer index and an increase in the percentage of ulcer inhibition, bio-reduced AgNPs decreased the adverse effect of ethanol-induced stomach injury. Reduced mucosal damage and hemorrhagic lesions, necrobiotic alterations in the stomach epithelium, submucosal edema, and blood vessel congestion were all indicators of substantially reduced ethanol-induced gastric lesions and greater anti-inflammatory results.

Conclusions: Fruit pellicle extract-derived bio-reduced silver nanoparticles have an efficacy similar to proton pump inhibitors like omeprazole and a lower incidence of possible side effects, making them a safe herbal therapy for ulcers.

Resumen

Contexto: Las úlceras pépticas constituyen un problema difícil. Las nanopartículas de plata (AgNPs) desempeñan un papel sustancial en las bioactividades y tienen características distintivas. La nanotecnología biorreductora tiene varios usos biomédicos. Históricamente, la atención sanitaria tradicional ha empleado Juglans regia L.

Objetivos: Evaluar la eficacia de las nanopartículas de plata biorreducidas (AgNP/FP) derivadas del extracto de la película de la fruta en el tratamiento de las úlceras pépticas.

Métodos: Para caracterizar las AgNP y las AgNP/FP se utilizaron espectros ultravioletas, dispersión dinámica de la luz, microscopía electrónica de transmitancia e infrarrojo transformado de Fourier. Las AgNPs y los extractos se compararon con el omeprazol en úlceras de estómago inducidas por etanol en ratones. Se crearon al azar seis grupos de 36 ratones. Se administraron AgNPs por vía oral a los grupos experimentales, utilizando 300 mg/kg como dosis. El inhibidor de la bomba de protones de referencia, omeprazol, se empleó como control.

Resultados: Como lo demuestra el descenso del índice de úlcera y el aumento del porcentaje de inhibición de la úlcera, las AgNPs biorreducidas disminuyeron el efecto adverso de la lesión estomacal inducida por etanol. La reducción del daño de la mucosa y de las lesiones hemorrágicas, las alteraciones necrobióticas del epitelio estomacal, el edema de la submucosa y la congestión de los vasos sanguíneos fueron indicadores de una reducción sustancial de las lesiones gástricas inducidas por etanol y de mayores resultados anti-inflamatorios.

Conclusiones: Las nanopartículas de plata biorreducidas derivadas del extracto de película de fruta tienen una eficacia similar a la de los inhibidores de la bomba de protones como el omeprazol y una menor incidencia de posibles efectos secundarios, lo que las convierte en una terapia herbal segura para las úlceras.
